Drug intelligence / Profile preview
SOCS1 peptidomimetic refers to a class of synthetic peptide-based therapeutics designed to mimic the kinase inhibitory region (KIR) of the endogenous Suppressor of Cytokine Signaling 1 (SOCS1) protein. SOCS1 is a key negative feedback regulator of the JAK/STAT signaling pathway, which is chronically activated in various inflammatory, autoimmune, and metabolic diseases. By mimicking the KIR domain, these peptidomimetics (such as MiS1 and PS5) bind to the catalytic domain of Janus kinases (primarily JAK1 and JAK2), acting as pseudo-substrates to inhibit their kinase activity and subsequent STAT1/3 phosphorylation. Preclinical studies, primarily led by researchers at the Fundacion Jimenez Diaz, Autonoma University of Madrid, and the University of Florida, have demonstrated that SOCS1 peptidomimetics can reduce inflammation, oxidative stress, and fibrosis in animal models of diabetic nephropathy, diabetic retinopathy, atherosclerosis, metabolic dysfunction-associated steatotic liver disease (MASLD), and uveitis.
