A TNF-neutralizing antibody is a biologic therapeutic designed to bind and inhibit tumor necrosis factor (TNF), a potent pro-inflammatory cytokine. By preventing TNF from binding to its receptors (TNFR1 and TNFR2), these antibodies disrupt inflammatory signaling cascades that contribute to various chronic diseases. Clinically, this class of drugs is a cornerstone in the management of chronic inflammatory conditions such as rheumatoid arthritis, Crohn's disease, and plaque psoriasis. Emerging research, such as that presented at AHA 2025, suggests that TNF-neutralizing antibodies may also play a role in reducing cardiovascular risk in cancer patients by mitigating a TNF-driven angiogenic pathway (involving LRG1) that accelerates atherosclerosis and plaque instability.
