Tofacitinib + R-ICE is a combination therapy consisting of tofacitinib, an oral small molecule Janus kinase (JAK) inhibitor, and the R-ICE chemotherapy regimen. Tofacitinib inhibits JAK1 and JAK3, interfering with the JAK-STAT signaling pathway involved in immune cell function and cytokine-mediated inflammation. The R-ICE regimen includes rituximab (a monoclonal antibody targeting CD20 on B cells), ifosfamide (an alkylating agent), carboplatin (a platinum-based chemotherapeutic), and etoposide (a topoisomerase II inhibitor). This combination is being investigated for use in relapsed or refractory diffuse large B-cell lymphoma, aiming to enhance anti-tumor efficacy by combining targeted immunomodulation with cytotoxic chemotherapy[2].
