This is a combination of two oral small molecule Janus kinase (JAK) inhibitors, tofacitinib and upadacitinib. Both drugs are used for the treatment of autoimmune and inflammatory diseases, particularly moderate-to-severe rheumatoid arthritis, psoriatic arthritis, ulcerative colitis, and other related conditions. Tofacitinib is a pan-JAK inhibitor with preferential selectivity for JAK1 and JAK3, while upadacitinib is a second-generation selective JAK1 inhibitor. By inhibiting these kinases, both drugs disrupt cytokine signaling pathways critical in the pathogenesis of chronic inflammation. The combination would theoretically provide broad inhibition across multiple JAK isoforms but is not an approved or standard therapy; each drug is typically used as monotherapy or in combination with other disease-modifying antirheumatic drugs (DMARDs), not together[2][7][9].
