**TT-00420 + atezolizumab** is a combination therapy of two agents: tinengotinib (TT-00420), a novel oral multi-kinase inhibitor, and atezolizumab, an anti-PD-L1 monoclonal antibody. - **Tinengotinib** targets multiple kinases implicated in cancer proliferation and immune regulation, including fibroblast growth factor receptors 1–3 (FGFR1, FGFR2, FGFR3), Janus kinase 1/2 (JAK1, JAK2), vascular endothelial growth factor receptors (VEGFRs), and Aurora kinases A/B. - **Atezolizumab** is a monoclonal antibody that inhibits programmed death-ligand 1 (PD-L1), a crucial immune checkpoint, thereby promoting anti-tumor immune responses. This combination has been investigated primarily in advanced biliary tract carcinoma (BTC) and cholangiocarcinoma, based on preclinical rationale for synergistic anti-tumor effects and observed clinical benefit in patients previously treated with immunotherapies. The combination is under development, notably evaluated in phase Ib/II trials in heavily pretreated solid tumor patients, with results indicating promising efficacy and tolerability[4].
