upadacitinib + topical corticosteroids (AbbVie Inc.)

Upadacitinib + topical corticosteroids is a combination therapeutic approach used for the treatment of moderate to severe atopic dermatitis in adults and adolescents. Upadacitinib is an oral, selective Janus kinase 1 (JAK1) inhibitor developed by AbbVie that modulates the signaling of various pro-inflammatory cytokines (such as IL-4, IL-13, and IL-31) involved in skin inflammation and pruritus. Topical corticosteroids (TCS) are anti-inflammatory agents that act by binding to the glucocorticoid receptor, leading to the downregulation of pro-inflammatory genes and the reduction of local skin inflammation. This combination was specifically evaluated in the pivotal Phase 3 AD Up clinical trial (NCT03568318), which demonstrated that the addition of upadacitinib to a background of TCS significantly improved skin clearance and reduced itch compared to TCS alone in patients who were candidates for systemic therapy.
