Overview

WTX-2022 is a systemically delivered, conditionally activated T-cell engager (TCE) molecule being developed by Werewolf Therapeutics using its proprietary INDUCER platform. It targets Cadherin-6 (CDH6), a tumor-associated antigen frequently overexpressed in ovarian and kidney cancers. The molecule is designed with a masking approach and novel linker technology that ensures it remains inactive in peripheral tissues, thereby addressing the systemic toxicity limitations of conventional T-cell engagers. Upon reaching the tumor microenvironment, the mask is selectively cleaved, allowing the antibody to engage T cells against CDH6-expressing tumor cells. Preclinical data have demonstrated strong tumor-specific activation, antitumor activity in xenograft models, and a favorable safety profile with low cytokine release in non-human primates.
