Overview

- ZG005 + gecacitinib is an experimental combination of two drugs: ZG005, a bispecific antibody that simultaneously targets PD-1 (Programmed cell death protein 1) and TIGIT (T cell immunoreceptor with Ig and ITIM domains), and gecacitinib, a small molecule inhibitor of Janus kinases (JAK1, JAK2, JAK3, and TYK2) and ACVR1. - ZG005 works by blocking both the PD-1 and TIGIT immune checkpoints, aiming to synergistically activate T cells and enhance anti-tumor immune responses. - Gecacitinib inhibits the JAK-STAT pathway and ACVR1, reducing inflammation, improving anemia by downregulating hepcidin, improving iron metabolism, and reducing splenomegaly. - Both drugs are under clinical development by Suzhou Zelgen Biopharmaceuticals, primarily for oncological and hematological indications such as advanced solid tumors and myelofibrosis[1][2][4][5][6].
