Overview

ZKY018 is an investigational topical gel formulation of tofacitinib, a small molecule Janus kinase (JAK) inhibitor, being developed by Lee's Pharmaceutical and its subsidiary Zhaoke Pharmaceutical (ZKGZ Biotech) for the treatment of inflammatory dermatological conditions. As a pan-JAK inhibitor, ZKY018 targets the Janus kinase family of enzymes (primarily JAK1, JAK2, and JAK3), thereby blocking the JAK-STAT signaling pathway which is a critical mediator in the production of pro-inflammatory cytokines. This mechanism is intended to reduce skin inflammation, redness, and pruritus (itching) associated with conditions such as atopic dermatitis. By utilizing a topical delivery system, ZKY018 aims to provide localized therapeutic effects while minimizing the systemic exposure and potential side effects typically associated with oral JAK inhibitors. The drug is currently in Phase 2 clinical development for the treatment of mild to moderate atopic dermatitis.
