teclistamab
Primary Analysis of the phase 3 randomized MonumenTAL-6 study of teclistamab (Tec) plus talquetamab (Tal) or Tal plus pomalidomide (Pom) vs investigator's choice of elotuzumab, Pom and dexamethasone (EPd) or Pom, bortezomib, and dexamethasone (PVd) in patients (pts) with relapsed/refractory multiple myeloma (RRMM) and 1–4 prior lines of therapy (LOT)
Read the abstract (opens in a new tab)Tec+Tal and Tal+Pom significantly improved OS vs EPd/PVd (HRs [95% CI]: 0.38 [0.23–0.60], P <0.0001 and 0.55 [0.36–0.83], P =0.0044, respectively).
ivosidenib
Superior event-free survival with ivosidenib plus intensive chemotherapy in newly diagnosed IDH1-mutated AML: Results from the phase 3 HOVON150/AMLSG29-18 trial
Read the abstract (opens in a new tab)Estimated 24-mo EFS was 74% (95% CI, 67–79) with IVO vs 35% (95% CI, 28–42) with PBO.
epcoritamab
Epcoritamab plus lenalidomide (E-Len) compared to rituximab plus gemcitabine and oxaliplatin (R-GemOx): Primary results from the phase 3 EPCORE DLBCL-4 study in relapsed or refractory (R/R) diffuse large B-cell lymphoma (DLBCL)
Read the abstract (opens in a new tab)E-Len delivered a statistically significant and clinically meaningful PFS improvement vs R-GemOx with an HR of 0.40 (95% CI 0.30–0.55; P <0.0001).
venetoclax
Venetoclax, homoharringtonine, aclarubicin, and cytarabine (VHAA) versus daunorubicin and cytarabine (DA) as induction therapy for young adults with newly diagnosed acute myeloid leukemia: Results from a Phase III randomized controlled trial
Read the abstract (opens in a new tab)With median follow-up 35.7 vs. 37.5 months, median EFS was not reached in the VHAA group vs. 23.8 months in the DA group; 3-year EFS was 69.7% (95% CI, 60.5–77.1) vs. 47.5% (95% CI, 34.3–59.6), with a significant HR of 0.474 (95% CI, 0.280–0.805; P=0.002).
vamotinib
Efficacy and safety of vamotinib versus high-dose imatinib in patients with chronic-phase CML resistant to standard-dose imatinib: 24-month Results of a multicenter randomized phase III study
Read the abstract (opens in a new tab)MR4 was achieved in 45% vs 14% (Hazard ratio 3.84; 95% CI 2.01–7.35; p<0.0001).
elranatamab
Elranatamab (ELRA) monotherapy versus daratumumab plus pomalidomide and dexamethasone (DPd) in patients with relapsed or refractory multiple myeloma (RRMM): Results from the phase 3 MagnetisMM-5 trial
Read the abstract (opens in a new tab)Median PFS was not reached (95% CI, 41.7-NE) for ELRA vs 27.1 (95% CI, 17.6-32.5) mo for DPd; 36-mo PFS rate was 63.5% (95% CI, 56.3-69.8) vs 41.1% (95% CI, 34.3-47.8) for ELRA and DPd, respectively.
zanubrutinib
Superior progression-free survival with zanubrutinib plus rituximab vs bendamustine-rituximab in treatment-naïve mantle cell lymphoma (TN MCL): Primary analysis of a phase 3 study (MANGROVE)
Read the abstract (opens in a new tab)OS, though immature at this analysis, was numerically superior with ZR over BR (HR: 0.71 [95% CI: 0.51-0.99]; 1-sided P= .0222) with 4-year OS rates of 75.1% vs 67.6%, respectively.
loncastuximab tesirine
Results from the Phase 3 LOTIS-5 study: Efficacy and safety of loncastuximab tesirine plus rituximab vs immunochemotherapy (R-GemOx) in patients with relapsed or refractory DLBCL
Read the abstract (opens in a new tab)Investigator-assessed PFS demonstrated an HR of 0.65 (95% CI 0.51-0.81; nominal P =0.0002; median 5.49 vs 3.02 months).
rezafungin
ReSPECT: Phase 3 multicentre, randomized, double-blind study of once-weekly rezafungin versus standard antimicrobial regimen to prevent invasive fungal disease in adults undergoing allogeneic haematopoietic stem cell transplantation
Read the abstract (opens in a new tab)Time to IFD or death (Day 120) favoured rezafungin (adjusted HR 0.729 [95% CI 0.412, 1.291]).
methotrexate
Final results from a phase 3 double-blind, randomized placebo-controlled study evaluating methotrexate in combination with corticosteroids as initial treatment of acute graft-versus-host disease.
Read the abstract (opens in a new tab)ORR at Day 28 was 71% versus 75% (p=0.66) and ORR at Day 56 was 65% versus 63% for methotrexate vs. placebo respectively (p=0.84).
