aGD2-SIRPα fusion antibody is a recombinant fusion protein designed for the treatment of neuroblastoma, a childhood malignancy characterized by high expression of the disialoganglioside GD2. The drug consists of an anti-GD2 monoclonal antibody fused to the extracellular domain 1 of signal regulatory protein alpha (SIRPα). This dual-targeting approach aims to simultaneously bind GD2 on tumor cells and block the CD47-SIRPα innate immune checkpoint, also known as the "don't eat me" signal. By locally restricting CD47 blockade to the tumor site, the fusion antibody minimizes the on-target off-tumor toxicities typically associated with ubiquitous CD47 expression on healthy cells. The molecule is equipped with a functional Fc-domain that interacts with Fcγ receptors to induce innate immune effector mechanisms, including enhanced phagocytosis by macrophages and NK cell-mediated killing of neuroblastoma cells.
