Overview

anti-hPD-1 refers to a class of monoclonal antibodies designed to target and inhibit the human Programmed Cell Death Protein 1 (PD-1) receptor. PD-1 is a key immune checkpoint protein expressed on the surface of activated T cells, B cells, and myeloid cells. Under normal physiological conditions, PD-1 interacts with its ligands, PD-L1 and PD-L2, to suppress T-cell activity and maintain peripheral tolerance, preventing autoimmunity. However, many tumors exploit this pathway by upregulating PD-L1 to evade immune surveillance. Anti-hPD-1 antibodies block the interaction between PD-1 and its ligands, thereby "releasing the brakes" on the immune system and restoring the ability of cytotoxic T cells to recognize and eliminate cancer cells. This therapeutic strategy has revolutionized oncology, leading to the approval of several major drugs (e.g., pembrolizumab, nivolumab) for a wide range of indications, including melanoma, non-small cell lung cancer, and gastric cancer. In preclinical research, the term is frequently used to describe antibodies used in humanized mouse models and patient-derived xenografts (PDX) to study immune-mediated tumor growth inhibition and mechanisms of drug resistance.
