Overview

CBD-CCL4 is a tumor-targeted recombinant fusion protein designed to enhance the recruitment of specific immune cells into the tumor microenvironment. It consists of the chemokine CCL4 (C-C motif ligand 4) fused to a collagen-binding domain (CBD) derived from the A3 domain of von Willebrand factor. By binding to the collagen exposed in the leaky vasculature and stroma of tumors, CBD-CCL4 localizes the chemokine within the tumor, thereby recruiting CD103+ dendritic cells and CD8+ T cells. This mechanism is intended to overcome the "cold" tumor microenvironment and synergize with checkpoint inhibitor immunotherapies, such as anti-PD-1 or anti-CTLA-4, to improve treatment outcomes in patients who are otherwise non-responsive.
