This is a combination regimen consisting of three agents: - **Cisplatin** is a platinum-based chemotherapy that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - **Figitumumab** is a fully human monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), inhibiting IGF signaling implicated in cancer cell proliferation and survival. Figitumumab was developed by Pfizer but its development and manufacture were discontinued after 2011 due to safety concerns and lack of efficacy[8][6]. - **Pemetrexed** is an antifolate antineoplastic agent that inhibits several folate-dependent enzymes involved in nucleotide synthesis, thereby blocking DNA replication and cell division. This combination has been investigated for use primarily in non-small cell lung cancer (NSCLC), leveraging the cytotoxic effects of cisplatin and pemetrexed with the targeted action of figitumumab against IGF-1R. However, clinical development was halted due to adverse outcomes associated with figitumumab[8].
