Overview

This is a combination therapy consisting of three drugs—cyclophosphamide, hydroxychloroquine, and prednisolone—used primarily in the management of autoimmune diseases such as systemic lupus erythematosus (SLE), lupus nephritis, and rheumatoid arthritis. Cyclophosphamide is an alkylating agent that suppresses immune cell proliferation by cross-linking DNA. Hydroxychloroquine is a disease-modifying anti-rheumatic drug (DMARD) with immunomodulatory effects, thought to interfere with antigen presentation and toll-like receptor signaling. Prednisolone is a synthetic glucocorticoid that exerts potent anti-inflammatory and immunosuppressive actions by modulating gene expression via the glucocorticoid receptor. The combination aims to achieve disease remission while minimizing toxicity compared to high-dose corticosteroid regimens alone[6][1]. This regimen has been shown effective in inducing remission in lupus nephritis and may be used for other severe or refractory autoimmune conditions.
