KB-436 is a trispecific VHH-based antibody developed by KisoJi Biotechnology for the treatment of small cell lung cancer (SCLC) and other advanced solid tumors. It is designed to simultaneously target Dopamine Receptor 2 (DRD2), Programmed Cell Death Protein 1 (PD-1), and CD47. DRD2 is a G protein-coupled receptor often overexpressed in SCLC and associated with tumor growth and stemness. The PD-1 module aims to restore T-cell function, while the CD47 module blocks the CD47-SIRPα "don't eat me" signal and recruits T cells to the tumor microenvironment. Preclinical data indicates that KB-436 suppresses tumor growth, inhibits metastasis, and enhances the efficacy of chemotherapy.
