Overview

OX40L-Ig is a recombinant fusion protein consisting of the extracellular domain of the OX40 ligand (OX40L; CD252) fused to the Fc region of an immunoglobulin (Ig), typically IgG. It functions as a potent agonist of the OX40 receptor (CD134), a member of the tumor necrosis factor receptor superfamily (TNFRSF) expressed primarily on activated T cells. By binding to OX40, OX40L-Ig provides costimulatory signals that enhance T cell proliferation, survival, and cytokine production, particularly promoting Th2 and Th17 helper T cell differentiation. In preclinical research, it is used to study immune responses in models of asthma, autoimmune diseases, and parasitic infections. While various OX40 agonists have entered clinical trials (e.g., MEDI0562, GSK3174998), OX40L-Ig specifically refers to the fusion protein construct often used as a research tool to mimic the natural ligand's activity.
