PMG-A9 is an orally bioavailable small molecule designed as a first-in-class tumor suppressor activator for the treatment of various cancers. Developed by Promedigen, the compound restores the function of a key tumor suppressor that is frequently inactivated in malignancies. PMG-A9 has been shown to potently downregulate major oncogenic drivers and resistance factors, including FoxM1, Cyclin D1, and c-Myc, while upregulating pro-apoptotic proteins such as PUMA. In preclinical models of colorectal cancer and hormone receptor-positive breast cancer, PMG-A9 demonstrated robust single-agent antitumor efficacy and synergistic effects when combined with standard-of-care agents like 5-fluorouracil, CDK4/6 inhibitors, and fulvestrant, suggesting its potential to overcome therapeutic resistance.
