Overview
Poseltinib + rituximab + lenalidomide is an investigational drug combination currently evaluated for the treatment of relapsed/refractory B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma (DLBCL)[4]. Poseltinib is an oral Janus kinase (JAK) inhibitor that targets multiple isoforms including JAK1 and JAK2. Rituximab is a monoclonal antibody against CD20, mediating cell death via antibody-dependent cellular cytotoxicity (ADCC) and direct apoptosis. Lenalidomide is an immunomodulatory agent that enhances T-cell and NK-cell activity, also inducing apoptosis in malignant cells through cereblon-dependent mechanisms and modulating cytokine production[3][4]. The rationale for combining these modalities is synergistic immune activation and tumor cell targeting, as shown in mechanistic and early clinical studies for similar regimens[4]. This regimen is being studied in a prospective, open-label, multicenter phase 2 trial.
