Sema6A-Fc is a recombinant fusion protein consisting of the extracellular domain of Semaphorin 6A (Sema6A) fused to the Fc region of an immunoglobulin. It is being investigated as a potential therapeutic for pulmonary hypertension (PH), a condition characterized by endothelial cell apoptosis and vascular structural reconstruction. Research indicates that Sema6A expression is significantly downregulated in the lungs of patients with idiopathic pulmonary arterial hypertension (iPAH) and PH due to left heart disease (PH-LHD). Sema6A-Fc acts as an agonist that restores signaling through the Plexin-A4/VEGFR2 complex, which is essential for maintaining endothelial cell survival and preventing apoptosis. By stabilizing this signaling pathway, Sema6A-Fc attenuates pulmonary vascular remodeling, reduces right ventricular systolic pressure, and improves right ventricular function in preclinical models of PH.
