Overview

TA99-TNFα is an immunocytokine consisting of the TA99 monoclonal antibody fused to tumor necrosis factor-alpha (TNFα). The TA99 antibody component specifically targets gp75 (also known as tyrosinase-related protein 1 or TYRP1), a glycoprotein highly expressed on the surface of melanoma cells. By delivering TNFα directly to the tumor microenvironment, the fusion protein aims to induce pro-inflammatory changes, increase the recruitment and activation of immune effector cells such as natural killer (NK) cells, and enhance antibody-dependent cell-mediated cytotoxicity (ADCC). Developed by researchers at ETH Zürich, TA99-TNFα has demonstrated the ability to inhibit tumor growth and potentiate the activity of intact IgG antibodies in preclinical melanoma models.
