Overview

TYRP1-TriNKET is a trispecific immune cell engager developed by Dragonfly Therapeutics using their proprietary TriNKET® platform. It is designed to target Tyrosinase-related protein 1 (TYRP1), a melanocyte protein expressed on melanomas and certain other tumors. The molecule simultaneously engages TYRP1 on tumor cells and the activating receptors CD16a and NKG2D on cytotoxic immune cells, including natural killer (NK) cells, γδ T cells, and CD8+ T cells. This multi-receptor engagement facilitates the recruitment and activation of these effector cells within the tumor microenvironment, leading to potent anti-tumor activity. Preclinical studies have demonstrated superior efficacy compared to monoclonal antibodies and broad combinability with checkpoint inhibitors and various chemotherapies.
