Overview

AD-109 is a fixed-dose triple combination therapy being developed by Addpharma, a subsidiary of Yuhan Corporation, for the treatment of type 2 diabetes mellitus. The drug candidate combines three established anti-diabetic agents with complementary mechanisms of action: empagliflozin (a sodium-glucose cotransporter 2 inhibitor), linagliptin (a dipeptidyl peptidase-4 inhibitor), and metformin hydrochloride (a biguanide). By utilizing three distinct pathways—promoting urinary glucose excretion, enhancing incretin-mediated insulin secretion, and reducing hepatic glucose production—AD-109 aims to provide superior glycemic control and improved patient compliance compared to individual or dual therapies. It is currently in Phase 1 clinical development in South Korea and is classified as an incrementally modified drug (IMD).
