Overview

Avelumab + xevinapant is an investigational combination therapy for cancer, consisting of two agents with complementary mechanisms. Avelumab is an anti-PD-L1 monoclonal antibody that blocks the interaction between PD-L1 and its receptor PD-1, thereby enhancing T-cell mediated antitumor immune responses. Xevinapant is an oral small molecule inhibitor of inhibitor of apoptosis proteins (IAPs), which restores apoptosis in tumor cells and can increase antitumor immunity by modulating immune signaling pathways. The combination has been evaluated in phase Ib clinical studies for advanced solid tumors and non-small cell lung cancer, aiming to enhance efficacy through both direct tumor cell killing and immunomodulation[1][2][3].
