Overview

This is a combination drug consisting of three active pharmaceutical ingredients: - **Codeine phosphate** is an opioid analgesic used for the relief of mild to moderate pain. It acts primarily as a prodrug, being metabolized in the body to morphine, which then exerts its effects by binding to mu-opioid receptors in the central nervous system, resulting in analgesia and cough suppression[5]. - **Prednisolone** is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive properties. It works by binding to glucocorticoid receptors, inhibiting pro-inflammatory gene expression and promoting anti-inflammatory pathways. Prednisolone also inhibits phospholipase A2 activity, reducing arachidonic acid derivatives and suppressing immune cell migration[4]. - **Sulphasalazine (sulfasalazine)** is a disease-modifying anti-rheumatic drug (DMARD) used mainly for inflammatory bowel diseases such as ulcerative colitis and Crohn's disease, as well as rheumatoid arthritis. Its mechanism involves inhibition of prostaglandin synthesis (via prostaglandin G/H synthase 1/2), antagonism of phospholipase A2, inhibition of nuclear factor kappa-B signaling pathways, and modulation of immune responses[1][2][6][7]. This combination may be used together for synergistic management of inflammatory conditions such as ulcerative colitis or Crohn's disease where pain control (codeine), inflammation reduction (prednisolone), and long-term immunomodulation (sulphasalazine) are required[1][3]. There is no evidence that this specific triple combination exists under a unique brand name; it appears to be prescribed off-label or compounded when clinically indicated.
