danazol + prednisolone

Danazol + prednisolone is a combination therapy consisting of two small molecule drugs: danazol, a synthetic steroid with antigonadotropic and anti-estrogenic properties, and prednisolone, a glucocorticoid corticosteroid. Danazol acts primarily by inhibiting pituitary gonadotropin secretion and has some androgenic effects. Prednisolone exerts potent anti-inflammatory and immunosuppressive actions through activation of the glucocorticoid receptor. This combination has been used as an alternative treatment for paroxysmal nocturnal hemoglobinuria (PNH), particularly in settings where standard therapies such as eculizumab or anti-lymphocyte globulin are unavailable or unsuitable. The regimen may also be considered in other hematological disorders where both immunosuppression and hormonal modulation are beneficial[1][4].
