OAd-NIS is an oncolytic adenovirus engineered with a ∆E3 structure to express the sodium iodide symporter (NIS) as a function of viral replication. Developed by researchers at the University of Minnesota, it features an Ad 5/3 chimeric fiber for optimized binding and transduction in colorectal cancer (CRC) cells and utilizes a Cox2 promoter to drive tumor-specific expression. As a theranostic agent, OAd-NIS enables both diagnostic imaging via radiotracer uptake (e.g., 124I for PET/CT) and therapeutic efficacy when combined with radioactive iodine (e.g., 131I). The virus selectively replicates in and lyses tumor cells while simultaneously concentrating radioactive iodine within the tumor mass, leading to enhanced tumor growth suppression.
