Overview

Sintilimab + anlotinib + taxane-based chemotherapy is an investigational combination regimen being evaluated for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC). The regimen combines three distinct therapeutic approaches: immune checkpoint blockade, anti-angiogenesis, and cytotoxic chemotherapy. Sintilimab is a fully human IgG4 monoclonal antibody that targets the PD-1 receptor, preventing its interaction with PD-L1 and PD-L2 to reactivate anti-tumor T-cell responses. Anlotinib is an oral, small-molecule multi-target tyrosine kinase inhibitor that suppresses tumor growth and angiogenesis by inhibiting VEGFR, FGFR, PDGFR, and c-Kit. The taxane component (typically paclitaxel or docetaxel) acts as a microtubule stabilizer, interfering with cell division and inducing apoptosis. This triple combination is primarily being studied in patients who have progressed after prior platinum-based chemotherapy or immunotherapy.
