TIM3-Fc is a recombinant fusion protein consisting of the extracellular domain of T-cell immunoglobulin and mucin-domain containing-3 (TIM-3) fused to an immunoglobulin Fc region. It acts as a decoy receptor or blocking agent for TIM-3 ligands, specifically Galectin-9. In the context of chronic graft-versus-host disease (cGVHD), TIM3-Fc disrupts the pathogenic crosstalk between Galectin-9+ senescent macrophages and TIM3+ stem-like memory CD8+ T cells (Tscm). By inhibiting this axis, it reduces the secretion of senescence-associated secretory phenotype (SASP) factors such as IL-6, IL-1β, and TGF-β, thereby ameliorating tissue fibrosis in organs like the lung and liver. The molecule is primarily used in preclinical research to investigate the role of the TIM3-Galectin9 axis in immune-mediated inflammatory and fibrotic conditions.
