Overview

TA99-IL12 is a preclinical-stage immunocytokine developed by researchers at ETH Zürich. It is a recombinant fusion protein comprising the TA99 monoclonal antibody, which specifically targets the tumor-associated antigen gp75 (also known as tyrosinase-related protein 1 or TYRP1), and the pro-inflammatory cytokine interleukin-12 (IL12). The gp75 antigen is highly expressed in both murine and human melanoma cells. The primary mechanism of TA99-IL12 involves the targeted delivery of IL12 to the tumor microenvironment, which recruits and activates immune effector cells, particularly natural killer (NK) cells and T cells. This activation is intended to potentiate antibody-dependent cell-mediated cytotoxicity (ADCC) against the tumor mass. In preclinical studies using B16F10 melanoma mouse models, TA99-IL12 has demonstrated the ability to inhibit metastasis and, when used in combination with other therapies, significantly reduce the growth of established subcutaneous tumors by increasing the density of immune effector cells within the neoplastic lesions.
